CHO stable-transfection serum-free cell culture media
Chinese hamster ovary (CHO) cells have become the gold standard for biopharmaceutical production and are currently the most widely used mammalian host, especially for the expression of human therapeutic proteins. It has been reported that approximately >70% of recombinant therapeutic proteins are produced in CHO cells. CHO cells are capable of more accurate protein glycosylation, assembly and folding like human cells. Therefore, for some proteins whose activity is closely related to post-translational modification, CHO cells become the best production host. CHO cells can also efficiently synthesize very large molecules that cannot be actively expressed in prokaryotic hosts, such as blood factors.
The earliest culture media for animal-derived cells consisted of components such as serum (e.g., fetal bovine serum), plasma, or tissue extracts, which provide vitamins, growth factors, and other nutrients needed for cell growth and proliferation, but its composition is complex and undefined, resulting in high batch variability and risk of contamination.
Media without supplementation of serum is a more preferred solution. Serum-free cell culture media are balanced solutions with a chemically defined nutrient composition which is precisely known and defined, ensuring lot-to-lot consistency, and eliminating the need to screen new serum lots. For biopharmaceutical manufacturers, who are committed to eliminating all animal-derived components from their manufacturing processes, it is now possible to completely switch to serum-free media solutions when it comes to cell culture media.
