Disc-shaped microcarrier is a kind of efficient microcarrier dedicated to the adhesion culture of mammalian cells, with single layer thickness of 0.44 mm and pore size of about 15 μm. Sufficient specific surface area can provide sufficient surface area for cell growth, allowing cells to expand and grow in a three-dimensional structural space, while maintaining good exchange of various nutrients, and reduce the accumulation of metabolic wastes, thus provide a good growth environment for cells, and achieve high cell density.
Disc-Shaped Microcarrier Manufacturing Process
The manufacturing process adopts modern advanced thermal welding technology in D-level cleanroom to prevent secondary contamination from affecting the quality of the products. There are no additives in the processing process, in accordance with GMP requirements, the manufacturing process is recorded in detail, so that the product can be traced to the raw material. Physical and chemical properties were tested for each batch of raw materials; and endotoxin, bioburden, and cytotoxicity of each batch of products were analyzed and reported. The raw material is made of high purity polyester fiber which is non-toxic to cell growth and meets pharmaceutical requirements.

Applications
The disc-shaped microcarrier is suitable for adhesion dependent cells such as mammalian cells and insect cells, and the products are mainly secreted proteins and viruses. It can be used in packed bed bioreactor, single-use bioreactor, and culture flasks.
These microcarriers have been widely used in vaccine production processes, such as human rabies vaccine, IPV, Japanese encephalitis vaccine, EPO, swine fever vaccine, pseudorabies vaccine, avian influenza vaccine. Cell lines that have been successfully cultured include VERO, CHO, BHK, MDCK, ST, MARC-145, MRC-5, SF9, HEK293, 123A, 127A, GAMMA and primary cells.
Recommended Use Protocol
The use of disc-shaped microcarrier generally follows the following process.
1. Soak in 0.1N NaOH for more than 4 hours to completely wet the microcarrier.
2. Rinse with purified water until the PH is close to the PH of purified water.
3. Load into the bioreactor, add PBS solution, sterilize for 45 minutes, the loading capacity is calculated at 35 grams per liter.
4. After cooling, remove PBS and add cell culture medium for sterility test for 48 hours.
5. Remove the medium for sterile test, add medium and seed cells, and the density of the inoculated cells is generally 0.3-0.6 x 10^6 per milliliter.
6. Start cell culture process.
